Emma Stone shared the victory with her other nominees and thanked them.
She congratulated Yorgos Lanthimos, the director of the movie, as well as the whole cast and crew.
Attributing it to Ryan Gosling's performance, Stone made fun of her damaged gown.
She gave a heartfelt thank you to her daughter in her acceptance speech.
Following Stone's victory, host Jimmy Kimmel joked about "Envelopegate," alluding to an earlier Oscars gaffe.
With her victory for "La La Land" in 2017, Stone has now won two Oscars for best actress.
Presenters of the honor included Charlize Theron and Jennifer Lawrence, past winners of outstanding lead actress.
After accepting the prize, Stone gave her pal Jennifer Lawrence a hug.
"Poor Things" won four Oscars, including best actress, and was nominated for eleven in all.
